determine how transcription and mRNA stability are altered in embryos from old females. Secondly, we will use ribosome profiling to determine how translation efficiency changes across the transcriptome in embryos from old vs young parents. Thirdly, we will validate and further study gene expression changes using state-of-the-art quantitative imaging approaches, including smFISH in combination with MS2 and SunTag live imaging. This will include testing whether mRNA localisation in P-bodies, phase separated compartments associated with mRNA storage and degradation, is disrupted in embryos from old females. Machine learning will be used to uncover genomic features associated with changes in transcription, mRNA stability or translation in the embryos from young vs old parents. These genomic features will be used to identify links with known regulatory proteins, to develop hypotheses about the mechanisms underpinning the gene expression changes. Overall, results from this project will reveal new insights into the molecular mechanisms associated with poorer embryo survival upon parental ageing. A longer term goal is to exploit the findings relating to gene expression changes to identify genetic strategies to improve embryo quality and hatching rates when they are laid from aged females.
Entry Requirements
Applicants are expected to hold (or about to obtain) a minimum upper second class undergraduate honours degree (or equivalent) in a biology related subject. Experience in developmental biology, working with Drosophila and/or microscopy is desirable.
